Temperature Induction of Large Scale Bacterial Cultures

1. Grow up 500 mL culture in LB + antibiotic at 30°C to OD600 0.6-0.8

2. Have equal volume of LB + antibiotic warmed to 54°C

3. Induce culture by adding 54°C media to cell culture

4. Grow culture at 42°C for 2-4 hrs depending on protein expression

5. Separate proteins on SDS-PAGE and analyze
